Weight change at 68 weeks
REDEFINE 5 · Estimated average weight reduction
Two signals, one controlled trial
Cagrilintide is an amylin analogue, while semaglutide acts at the GLP-1 receptor. REDEFINE 5 randomly assigned 331 adults in Japan and Taiwan to a fixed cagrilintide-semaglutide combination or semaglutide alone, with lifestyle intervention in both groups. The 68-week study was double-blind and phase 3a. It tested a defined investigational product in a defined population, not the general idea of mixing two peptides. [1]
What the human result says
Estimated average body-weight change at week 68 was 18.4% lower with the combination and 11.9% lower with semaglutide alone, a difference of 6.5 percentage points. Adverse events were reported by 87% and 84% of participants, respectively, and gastrointestinal disorders were the most common. The study was funded by Novo Nordisk. These results support the tested combination under trial conditions; they do not establish equivalence for a compounded or research-labeled product. [1]
What the animal and laboratory work adds
A separate 2026 study mapped more than 530,000 cells from rat, mouse and macaque brainstem tissue and used rat experiments to investigate neural pathways involved in cagrilintide's effects on energy balance. The atlas found related cell populations across species, including a conserved population described in humans. However, the causal intervention experiments were performed in rats. This work helps explain a possible mechanism; it is not a human weight-loss trial. [2]
The label-matching check
Before connecting a study to a product, match the exact ingredients, their amounts and ratio, the finished formulation, manufacturer, route and storage instructions. A vial that simply lists cagrilintide and semaglutide has not automatically reproduced the trial product. Signal Axiom can help explain evidence and calculate from confirmed label information, but arithmetic cannot verify identity, sterility, purity or clinical suitability.
What remains unknown
REDEFINE 5 does not answer whether every population will have the same results, what happens beyond 68 weeks, or whether an independently produced blend matches the studied product. It also does not create a do-it-yourself protocol. Product selection and treatment decisions belong with a licensed clinician using verified product information.

Before applying a study to yourself, what should you check?
Check who or what was studied, which product was used, what it was compared with, and what actually changed. A laboratory finding or a change in a measured marker does not automatically establish a benefit for people. Use the sources below to check those details.
